🧬 EWING FAMILIES: ASK ABOUT STEAP1 🧬

A STEAP1-targeted clinical trials are open now, with additional STEAP1 cellular therapies in development.

STEAP1 is one of the most important immune targets we are watching in Ewing sarcoma right now.

Why?

Because STEAP1 is present on the surface of the vast majority of Ewing tumors, and recent research found it in about 97% of primary Ewing samples studied. Its expression is also linked to EWS::FLI1, the fusion protein that drives most Ewing sarcomas.

That gives researchers something extremely valuable:

A target on the outside of the Ewing cell that immune therapies can potentially lock onto.

And right now, there are two separate STEAP1 strategies families should know about.

⸻

1️⃣ AVAILABLE NOW: STEAP1 × CD3 T-CELL ENGAGER

The therapy is called xaluritamig.

It is designed to grab:

STEAP1 on the Ewing cell
and
CD3 on the patient’s own T cell

bringing the immune cell directly to the cancer.

It is an engineered T-cell engager that redirects the patient’s existing T cells toward STEAP1-positive tumor cells.

Current trial that is enrolling or waitlisting based on available spots:

NCT07297979
Phase 1b
Relapsed/refractory Ewing sarcoma
Age 2+

The current registry lists 11 recruiting sites, including eight in the U.S. and three in Australia.

U.S. recruiting sites 🇺🇸

Cedars-Sinai Medical Center — Los Angeles, CA
UCLA — Los Angeles, CA
Johns Hopkins University — Baltimore, MD
Dana-Farber Cancer Institute — Boston, MA
Mayo Clinic Children’s Center — Rochester, MN
Memorial Sloan Kettering — New York, NY
Children’s Hospital of Philadelphia — Philadelphia, PA
MD Anderson Cancer Center — Houston, TX

Australia 🇦🇺

Chris O’Brien Lifehouse — Sydney
Peter MacCallum Cancer Centre — Melbourne
Perth Children’s Hospital — Perth

⸻

📞 HOW FAMILIES CAN CONTACT THE TRIAL

Amgen Trial Information
📞 866-572-6436
📧 [email protected]

Or Clinicaltrials. Org for specific contacts to each location (located at the bottom of the trial link)
https://clinicaltrials.gov/study/NCT07297979

Families can ask Amgen which site has the correct age/dose cohort open now and whether another center has a faster opening.

So when calling, ask:
“Is there a slot available?”
“Is there a waitlist?”
“When is the next slot expected?”
“Does another site have availability sooner?”

⸻

2️⃣ WHAT’S COMING NEXT: STEAP1 CAR-T

This is the part XIT is watching extremely closely.

Researchers led by John Lee, MD, PhD, at UCLA have developed genetically engineered STEAP1 CAR-T cells for Ewing sarcoma.

Then they added another layer:

IL-18 “armoring”

The CAR-T cells are engineered not only to recognize STEAP1 but also to release IL-18, an immune-signaling molecule intended to make the cells more potent inside the hostile environment of a solid tumor.

🐁 WHAT DID THE MOUSE STUDIES SHOW?

In preclinical Ewing models:

* High-dose STEAP1 CAR-T caused complete tumor regression *** this is very exciting!
* In disseminated disease, responses were dose-dependent
* IL-18–armored STEAP1 CAR-T produced complete responses in about 80% of mice at a lower CAR-T dose
* No overt toxicity was reported in those experiments

⚠️ These are mouse results only.
They are not pediatric response rates, and the study is currently a preprint, not yet peer-reviewed.

⸻

🧬 HOW CLOSE IS THE CAR-T TO CHILDREN?

This is what makes the program particularly interesting.

That is not an open Ewing CAR-T trial yet.

But this is much further along than simply having promising mouse data.

⸻

🔭 WHY XIT IS WATCHING STEAP1 SO CLOSELY

STEAP1 checks several boxes we look for in a pediatric cancer target:

✓ Highly expressed across Ewing tumors
✓ Located on the cell surface
✓ Connected to core Ewing biology
✓ Already actionable through a human trial today
✓ Multiple immune-engineering strategies can target it
✓ A CAR-T program is moving toward clinical translation

And importantly, the two approaches complement one another.

RIGHT NOW:

Xaluritamig uses the patient’s existing T cells.

WHAT MAY BE NEXT:

STEAP1 CAR-T genetically reprograms T cells to recognize the tumor and potentially stay active longer.

For families, the takeaway is simple:

STEAP1 is no longer just a laboratory target.

There is an Ewing trial available now — and a genetically engineered CAR-T program moving behind it.

That is why XIT believes STEAP1 is one of the most important Ewing targets to watch right now. 🧬🎗️

WHY XIT GOT INVOLVED. 🧬

Our involvement in this project began when we learned about Sid Sijbrandij, an entrepreneur diagnosed with osteosarcoma, who took an extraordinary approach to his own cancer treatment.

He went “founder mode” on cancer, bringing together scientists, researchers, and emerging technologies to develop a highly personalized treatment strategy, including cancer vaccines.

We watched him speak about his experience and found ourselves asking one question:

If a someone with the resources to accelerate his own treatment can do this, why can’t we create those same opportunities for children in our community?

We later learned about his support for Serova Bio and were particularly impressed by the scientific and medical expertise behind its work.

Serova Bio was co-founded by Michael E. Bryan, an MD-PhD researcher at the University of Oxford and affiliate scientist at the Broad Institute of MIT and Harvard.

The team also includes Dr. Jie Min (James) Lam, Chief Medical Officer, who studied medicine at the University of Cambridge and University College London Medical School.

What impressed us was not simply the scientific expertise behind the technology, but the team’s willingness to explore ways to bring emerging therapies to patients sooner.

Then something remarkable happened.

Families from our childhood cancer community, including parents of children with ewing sarcoma and osteosarcoma, began reaching out to XIT, asking whether we could help their children access this therapy.

And we knew this was exactly the kind of work XIT was created to do.

One of the greatest challenges in pediatric oncology isn’t simply discovering promising therapies. It’s getting them to children in time.

Traditional drug development and large clinical trials can take many years. For children with aggressive cancers, those years are often years they do not have.

What makes this collaboration particularly meaningful is Serova Bio’s willingness to provide access for children now.

With personalized vaccine manufacturing potentially taking approximately 12 weeks, this represents an opportunity to work on a timeline very different from conventional timelines.

These vaccines remain investigational. We do not yet know which children will benefit, or to what extent. But we believe that responsibly expanding access to promising science, while collecting and sharing clinical outcomes, is how progress can accelerate.

Serova Bio has proposed making personalized vaccine development available for up to 20 children through a $200,000 funded cohort.

For perspective, personalized treatment development can otherwise carry costs approaching that amount for a single patient.

Our goal at XIT is to help make this opportunity accessible to children who may benefit, without placing the financial burden on their families.

This is more than a fundraising initiative.

It is an opportunity to bring together scientists, oncologists, families, foundations, and donors to explore a different model for developing and accessing pediatric cancer therapies.

One that moves faster. One that learns from every patient. One that puts children at the center of innovation.

If you would like to help make this possible, you can donate at XIT.org, share this initiative with a foundation or potential donor, or organize a fundraiser within your own community.

This model also creates the opportunity to establish additional cancer-specific cohorts, expanding access to personalized vaccines across pediatric malignancies.

If your community or foundation would like to fundraise for a specific cancer cohort, please reach out to XIT! Together, we can help bring these promising therapies to more children.

Children deserve cures. And they deserve access to innovation now.

WOW.🤍

We are truly overwhelmed with gratitude for the Crockett family, who opened their home, brought their community together, and raised $7,830 for XIT.

They shared the importance of pediatric cancer awareness. They started conversations. They brought more people into a mission that our children desperately need the world to care about.

And together, their community turned that awareness into action.

Their fundraiser will help begin building XIT’s very first pipeline to help children access promising therapies. 🧬

To the Crockett family and everyone who gathered, donated, listened, and learned that evening:

Thank you for making our children matter in your community.
Thank you for helping us build what comes next.

We are so, so grateful, Dione Crockett 💛

🧬COMPLETE REMISSION IN ADVANCED WILMS TUMORA 17-year-old with nephroblastoma/Wilms tumor had battled cancer for 10 years...

🧬COMPLETE REMISSION IN ADVANCED WILMS TUMOR

A 17-year-old with nephroblastoma/Wilms tumor had battled cancer for 10 years and ultimately developed extensive treatment-resistant disease involving the abdomen, lungs, liver, pelvis and brain. No curative treatment options remained.

Tumor sequencing revealed PRAME, an intracellular cancer antigen.

Researchers collected his own T cells and genetically reprogrammed them with a PRAME-specific T-cell receptor (TCR) so they could recognize PRAME presented by HLA-A*02:01 and attack the cancer. The genetic instructions were delivered using the IMA203CD8 lentiviral vector.

What happened?

🔬 Day 9: biopsy showed massive engineered T-cell infiltration and tumor-cell death.

🔬 3 months: repeat biopsy found no viable tumor cells.

🔬 Day 200: measurable tumors had shrunk 69%, while testing showed molecular, metabolic AND histologic complete remission — meaning PET activity, tumor DNA and viable cancer on biopsy were no longer detectable.

At the August 2026 report, almost one year after the single T-cell infusion, imaging and blood testing still showed no evidence of active cancer, and the patient had returned to cycling and normal daily life.

🧬 XIT DECIPHERS THE SCIENCE

CAR-T generally recognizes proteins sitting on the outside of cancer cells.

TCR-T can go deeper.

Cells chop up proteins from inside themselves and display fragments on HLA molecules. These engineered T cells were programmed to recognize a PRAME fragment and kill the cell displaying it.

That opens the possibility of targeting cancer proteins CAR-T simply cannot see.

Why XIT is watching

Researchers have now analyzed tumors from 2,500+ pediatric patients and report PRAME across multiple high-risk childhood cancers.

KiTZ Heidelberg is preparing PRAMEtime, a Phase I/II study planned for 2027, with up to 18 children and adolescents ages 8–17 with PRAME-positive solid tumors.

⚠️ Important: this is one patient, treated through compassionate use — not a response rate and not proof of cure. Cytokine-release syndrome occurred and required treatment.

But for a metastatic, therapy-resistant childhood solid tumor to reach molecular + metabolic + histologic remission after genetically engineered T-cell therapy is a signal worth following closely.

🧬 ALL | ONE INFUSION. THE BODY BECOMES THE FACTORY.

Early human gene-therapy data: 3/3 MRD-negative complete responses

This one is fascinating.

VNX-101 is an experimental gene-delivery therapy being studied in people with relapsed or refractory CD19-positive blood cancers, including B-cell acute lymphoblastic leukemia (ALL).

And instead of repeatedly giving a cancer-fighting drug, researchers are trying something very different:

Give the body the genetic instructions once — and let the body keep making the therapy itself.

🔬 WHAT HAPPENED?

Among the first 3 evaluable patients with relapsed/refractory ALL who were negative for antibodies against the AAV vector before treatment:

3/3 achieved complete remission.

3/3 became MRD-negative.

All three MRD-negative results were confirmed using clonoSEQ, a highly sensitive test capable of detecting tiny amounts of residual leukemia.

One patient also had extensive leukemia outside the bone marrow and achieved a complete response at all disease sites on PET imaging by Day 28.

🧬 XIT DECIPHERS THE SCIENCE: HOW DOES ONE DOSE KEEP WORKING?

VNX-101 uses an engineered AAV — adeno-associated virus — as a delivery vehicle.

Think of the AAV as a microscopic delivery package.

Inside are genetic instructions telling primarily the patient’s liver cells how to manufacture a therapeutic protein called GP101.

GP101 is a CD19 × CD3 T-cell engager.

One end recognizes CD19 on leukemia cells.
The other recognizes CD3 on the patient’s T cells.

It essentially brings the patient’s own immune cells face-to-face with the leukemia and tells them:

“This is the cell to attack.”

But here’s the part we’re particularly interested in:

The liver keeps producing the cancer-targeting protein after the original infusion.

The first reported patient remained MRD-negative through Day 260 and then proceeded to stem-cell transplantation while still in MRD-negative remission.

Even after transplant, GP101 remained detectable at therapeutic levels at least 9 months after the original single VNX-101 infusion.

So rather than:

drug → drug disappears → give another dose

the concept is:

ONE INFUSION

↓
deliver genetic instructions
↓
the body becomes a long-lasting “factory”
↓
continuously produces the T-cell engager
↓
keeps recruiting T cells against CD19+ leukemia

That is why we’re watching this.

🎗️ WHAT ABOUT CHILDREN?

The 3 patients in this new report were adults — these are NOT pediatric results.

The current SENTRY-CD19 trial is a Phase 1/2 first-in-human study.

Part 1: ages 18–90

Part 2: the protocol allows expansion to patients 13–90 years old after dose selection and safety review.

VNX-101 has received FDA Rare Pediatric Disease, Fast Track and Orphan Drug designations. Those designations can facilitate development, but they do not mean the therapy is FDA-approved or currently available to children.

⚠️ WHAT FAMILIES SHOULD KNOW

This is very early evidence.

Three out of three MRD-negative complete responses is an exciting signal, but three patients cannot tell us what the eventual response rate will be.

These results were announced by the company and have not yet been published as peer-reviewed clinical results.

The first patient also proceeded to stem-cell transplantation. We therefore cannot say VNX-101 produced a cure or attribute subsequent long-term disease control solely to the gene therapy.

And because this is AAV-based gene delivery, patients must also be screened for antibodies against the AAV capsid. The current protocol requires an AAV antibody level below 1:400.

The trial includes long-term safety monitoring for 6–15 years, which is appropriate for a gene-delivery therapy designed to produce prolonged biological effects.

🔭 WHY XIT IS WATCHING

We’re interested in more than the 3/3.

We’re watching the platform.

If researchers can safely give the body genetic instructions that allow it to manufacture a therapeutic protein for months — rather than manufacturing personalized cells outside the body or repeatedly administering a drug — that could represent an entirely different way of delivering cancer immunotherapy.

And the concept may eventually extend beyond CD19 and ALL.

The underlying scientific question is enormous:

Can we program the body once and turn it into its own long-lasting medicine factory?

For now, we have an intriguing first human signal.

One infusion.
Three ALL patients.
Three MRD-negative complete remissions.
Therapeutic protein detected 9+ months after treatment in the first reported patient.

Now we need more patients, longer follow-up, detailed safety data, peer-reviewed clinical results — and pediatric data.

🧬 Very Exciting ALL News — ANNOUNCED TODAY

ONE IV DOSE → 3/3 MRD-NEGATIVE COMPLETE RESPONSES

September 21, 2026: New early human results were announced today from VNX-101, an investigational gene-delivered immunotherapy being studied in patients with relapsed/refractory acute lymphoblastic leukemia (ALL).

The first ALL results are small — but noteworthy:

3 patients treated.
3 MRD-negative complete responses.
All 3 confirmed by clonoSEQ.

One patient with extensive disease outside the bone marrow also achieved a complete response at all disease sites on PET by Day 28.

The first treated patient remained MRD-negative through Day 260 and then proceeded to stem-cell transplant while still in complete remission.

🧬 WHAT MAKES THIS DIFFERENT?

VNX-101 isn’t traditional CAR-T.

It uses an AAV vector to deliver genetic instructions in vivo. After a single IV administration, the patient’s liver is designed to continuously produce a CD19×CD3 T-cell engager, which brings the patient’s own T cells into contact with CD19-positive cancer cells.

In simple terms-

Instead of manufacturing cancer-fighting cells outside the body, this approach gives the body genetic instructions to continuously make a cancer-targeting therapy itself.

The platform is designed to avoid leukapheresis, ex-vivo CAR-T manufacturing and repeat therapeutic infusions.

🎗️ WHAT ABOUT CHILDREN WITH ALL?

This is particularly interesting for pediatric cancer because VNX-101 has already received FDA Rare Pediatric Disease designation, as well as Fast Track and Orphan Drug designations.

However, there is an important distinction for families:

Vironexis currently describes the actively enrolling VNX-101 trial as enrolling adults.

So this is not currently a treatment that every child with ALL can sign up to receive.

For families with a child or adolescent with relapsed/refractory CD19-positive ALL, it may still be worthwhile to have the treating oncologist contact the study team to ask about future pediatric enrollment and upcoming cohorts.

🔎 HOW TO FIND THE TRIAL

https://clinicaltrials.gov/study/NCT06533579

Families should contact the trial through the study contact information listed on ClinicalTrials.gov or have their oncologist contact the nearest participating center. The study team performs the actual prescreening and determines eligibility.

⚠️ IMPORTANT CONTEXT

3/3 MRD-negative complete responses is an encouraging early human signal — but three patients is still only three patients.

This is an early Phase 1/2 study, not evidence of a 100% response rate in a larger population and not evidence of a cure rate.

One of the patients subsequently underwent stem-cell transplantation, which also means longer-term remission after transplant cannot be attributed to VNX-101 alone.

Why XIT is watching 🧬

The concept is what makes this particularly compelling:

One IV administration → genetic instructions delivered inside the body → the patient’s own cells continuously manufacture a cancer-targeting immunotherapy.

If these early responses prove reproducible and durable in larger studies, this could represent an important new way to deliver cancer immunotherapy.
