Merck Announces Late-Breaking Data from Three Phase 3 Trials Evaluating Doravirine/Islatravir (DOR/ISL), an Investigational, Once-Daily, Two-Drug Regimen for the Treatment of Adults Living with HIV-1 at CROI 2026

DOR/ISL is the first non-INSTI, two-drug regimen to demonstrate non-inferiority and a similar safety profile at Week 48 to BIC/FTC/TAF in adults living with HIV-1 who had not previously received antiretroviral treatment DOR/ISL maintained virologic suppression at Week 96 in adults with virologically...

Amy Colson, MD, MPH, discusses data from the MK8591A-052 trial which showed that switching to doravirine/islatravir maintained durable virologic suppression with efficacy and safety comparable to continuing bictegravir/emtricitabine/tenofovir alafenamide.

NEW: A phase 2 study from Gilead Sciences and Merck finds that once-weekly oral islatravir plus lenacapavir (ISL+LEN) maintained high rates of virologic suppression at 48 weeks in people with HIV-1 with no serious adverse events. These results suggest that ISL+LEN may offer a promising once-weekly treatment regimen that could help address adherence challenges in HIV care. https://bit.ly/4p6X5Ek

"These results provide reassurance that the transition from daily to once-weekly dosing does not have an adverse effect on adherence and offers a favorable outlook for long-acting treatment strategies," Colson and colleagues wrote. "Offering a range of treatment options with differing administration routes and dosing intervals remains important to meet the diverse needs of persons with HIV-1 and minimize barriers to antiretroviral therapy adherence."

Access to prevention remains a cornerstone of our work. This year, with your help, CRI expanded our outreach and education efforts, ensuring that more people have information about and access to effective prevention. These initiatives are vital in reducing new infections and promoting overall community health.

As we face new challenges and shifting federal funding, your unrestricted support is crucial. On Giving Tuesday, we invite you to make a year-end gift to CRI. Your generosity will ensure that everyone, regardless of circumstance, receives the care and dignity they deserve. Together, we can build a future free from HIV, full of hope and opportunity.

As we mark World AIDS Day on December 1st, we pause to honor the resilience and courage of everyone affected by HIV in Massachusetts and beyond. At CRI, this day renews our commitment to ending stigma, supporting those in need, and advancing health equity for all. Thanks to your continued generosity, our community is stronger and more hopeful than ever before.

Today, CRI team members—Gerry, Wilfredo, and Steve—are representing our organization at the World AIDS Day event at Cyclorama. Yesterday, Steve joined the Ryan White Planning Council and helped carry the commemorative quilt from City Hall Plaza, honoring those we’ve lost and the ongoing fight against HIV.
