Pink oyster mushrooms (Pleurotus djamor) are edible mushrooms known for their bright pink color, attractive appearance, and delicate, slightly seafood-like flavor. They are rich in protein, dietary fiber, vitamins, minerals, and antioxidants, making them a nutritious food source. These mushrooms grow quickly on agricultural wastes such as straw, sawdust, and sugarcane bagasse, making them an environmentally friendly crop. However, they have a short shelf life and are best consumed soon after harvesting.

Tissue culture is a method of growing mushroom cells or tissues under sterile laboratory conditions to produce pure and healthy cultures. In pink oyster mushrooms, a small piece of tissue is taken from the inner part of a fresh fruiting body and placed on a nutrient medium, such as potato dextrose agar (PDA). The tissue develops into mycelium, which can be used to prepare spawn for commercial cultivation. Tissue culture helps maintain desirable characteristics, reduces contamination, preserves high-quality strains, and ensures uniform mushroom production throughout the cultivation process.

Mushroom spore culture is the process of growing mushrooms from spores under sterile laboratory conditions. Spores are the microscopic reproductive cells of mushrooms, similar to seeds in plants. They are collected from a mature mushroom using a spore print and transferred onto a sterile nutrient medium, such as agar, where they germinate and produce fine thread-like structures called mycelium. Grain Spawn to Commercial Spawn!
One important note is that repeatedly expanding grain spawn (grain-to-grain transfers) eventually increases the risk of contamination and can reduce culture vigor. Commercial producers typically limit the number of expansions by returning to a preserved master culture rather than continuously transferring from one grain batch to the next.

Making straw mushroom spawn bags involves preparing a clean, nutritious substrate using chopped straw or other agricultural residues. The substrate is moistened to the correct moisture level, packed into heat-resistant polypropylene bags, and sterilized or pasteurized to eliminate harmful microorganisms. After cooling, the substrate is inoculated with high-quality straw mushroom spawn under clean and hygienic conditions. The bags are then sealed and incubated in a warm, humid environment with proper ventilation. During incubation, the mushroom mycelium spreads throughout the substrate, producing healthy spawn bags that can later be used for successful straw mushroom cultivation.

Mushroom spawn bottles are sterilized in a metal drum to eliminate bacteria, fungi, and other harmful microorganisms before inoculation. The bottles filled with prepared substrate are arranged inside the drum on a rack to prevent direct contact with the bottom. Water is added to produce steam, and the drum is tightly closed. The bottles are sterilized at approximately 121°C under pressure, or by steaming for several hours if using a simple drum without pressure. After sterilization, the bottles are allowed to cool completely in a clean environment before inoculation with mushroom culture. Proper sterilization ensures healthy spawn growth and reduces the risk of contamination.
