JUST ANNOUNCED: The FDA has granted Breakthrough Therapy Designation to Servier Pharmaceutical’s Emi-Le—a B7-H4-targeting antibody drug conjugate—for ACC patients with solid histology or more aggressive forms of the disease. Breakthrough Therapy Designation is granted when early clinical data suggest a drug may provide substantial clinical benefit for a serious disease, particularly when treatment options are limited. The designation allows for closer collaboration with the FDA and may help accelerate the development and review process for the therapy.

Early results from the Emi-Le clinical trial presented in June of 2025 showed that 56% of ACC patients with aggressive disease had their tumors shrink by 30% or more.

For the 17th year, ACCRF will be participating in the American Society of Clinical Oncology (ASCO) annual meeting in Chicago this May which brings together leading oncologists from around the world to discuss the latest in cancer research and treatment. This year’s conference will feature presentations of new clinical data for two MYB degraders (REM-422 and RGT-61159) from biopharma companies Remix Therapeutics and Rgenta Therapeutics. ACCRF is proud to have collaborated with both these firms, providing preclinical drug screening data from our models that supported the opening of the clinical trials. ACCRF awards Dr. Piero Dalerba (Hackensack Meridian School of Medicine) a full grant to study "Mechanistic significance and therapeutic eradication of ductal and myoepithelial lineages in human Adenoid Cystic Carcinomas (ACCs)". This work seeks to provide a better understanding of a distinctive biological feature of ACC: that malignant tissue can contain two distinct sub-types of malignant cells (myoepithelial and ductal). The research will also test how effective current ADCs are at targeting both cell types in ACC tumor models.

A new phase 1 clinical trial for Next Cure's B7-H4 ADC (LNCB74) is now recruiting ACC patients across 14 US sites. This is an open-label, phase 1, dose escalation and dose expansion study to determine safety and tolerability, and to determine the maximum tolerated dose and / or recommended phase 2 dose of LNCB74 in participants with advanced solid tumors.

Congratulations to ACCRF Scientific Advisory Board Member Dr. Robert H. Vonderheide (UPenn Abramson Cancer Center) on his election as the new President of the American Association for Cancer Research (AACR). Dr. Vonderheide is an internationally renowned cancer immunotherapy and translational research expert whose work has uncovered the mechanisms of cancer immune surveillance and developed novel immunotherapeutics—such as vaccines, antibody-based therapies, and adoptive T-cell therapies. His expertise and tireless advocacy have helped guide ACCRF’s research efforts. As AACR President, Dr. Vonderheide will succeed Dr. Lillian Siu, who served on the ACCRF Scientific Advisory Board from 2011-2019 and is now an Emeritus Member. The ACC patient community is so fortunate to have benefited from the guidance of these world-leading oncologists.

Remix Therapeutics recently received FDA Fast Track designation for its drug REM-422, which targets MYB mRNA. Fast Track designation speeds up the development and review of drugs for serious diseases, especially when there are limited treatment options. Drugs in this program get more frequent communication with the FDA and results from their clinical trials may be reviewed faster.

REM-422 is currently being tested in a clinical trial recently named as ARIA (A Study of REM-422 In Adenoid Cystic Carcinoma). Early results presented in October showed that 43% of biomarker-positive ACC patients who took the drug for 6 months or longer had their tumors shrink by 30% or more.
