N of One was founded by parents and researchers who believe that breakthroughs in autism research that will lead to treatments and prevention strategies are possible in our lifetime. Our goal, simply put, is to identify the fundamental biological mechanisms of autism. Autism is now affecting 1 in 68 children and 1 in 42 boys in the US, that's almost 2.5%! In spite of this alarming rise and after y

ears of research and hundreds of millions of dollars of funding:

-We still don't even know what autism is
-We don't know what causes it
-We don't know how to prevent it
-And we don't have any treatments

Parents are understandably frustrated with this lack of progress, indeed we all should be. At N of One we have a view that things can be done better and look to partner with parents and researchers who share that view.

Leucovorin (Folinic Acid) has been in the news recently as a possible treatment for autism, however this is not new. It has been used by some doctors for years. Jim Simon's contribution to autism research is without parallel. He spent billions working to unravel the underpinnings of autism and leaves behind an institution that continues to support programs and researchers working to that goal.

I will always remember him for his kindness and inviting N of One to come share ideas about how to accelerate progress in autism research.

While there has been growing evidence of a link between Autism and the gut microbiome over the years, there have occasionally been researchers who suggest that the link wasn't there.

Recently a group of brilliant researchers re-analyzed the naysayers work and found that mistakes were made, and validated a strong link seen across dozens of studies.

Importantly, this work was funded by the Simon's Foundation, the largest private funder of ASD research in the world.

A SFARI initiated and funded reanalysis of previous studies reveals consistent biological signals in the human microbiome and other physiological signals associated with autism and highlights the need for long-term studies to determine autism’s underlying causes.

New N of One Supported Research Points Toward Three Autism Sub-groups with Distinct Prenatal Risk Factors

“Autism has always been diagnosed as one condition.” said Dr. Hahn, the study's author, “Now this work not only shows that there are subgroups of children with different co-occurring conditions, but that there are different prenatal risk factors associated with these subgroups. may not be sufficient in the future.”

The research found that infections, anti-inflammatory, and other complex medications were associated with one of the groups while immune disregulatory conditions such as asthma and joint disorders were associated with the second group, and lastly, overall pregnancy complications were associated with the third group of children.
