Australian researchers may have developed a therapeutic to impede sepsis from killing patients by preventing the cytokine storm that causes organ failure — survival rates jumped from 40% to 89%.

Sepsis — the body's catastrophic immune overreaction to infection — kills 11 million people annually, making it the leading cause of death in hospital intensive care units globally. The paradox of sepsis is that patients rarely die from the infection itself; they die from their own immune system's response, which triggers a cytokine storm that simultaneously causes uncontrolled clotting in small blood vessels, massive fluid leakage from capillaries, and multi-organ shutdown.

Researchers at the Walter and Eliza Hall Institute developed AntiSepsin — a monoclonal antibody targeting the HMGB1 protein, a late-stage inflammatory mediator released by dying immune cells that amplifies cytokine storm cascades. Unlike early-stage cytokine blockers that impair the patient's ability to fight infection, HMGB1 inhibition interrupts the damaging amplification loop that begins 16-32 hours into sepsis — after the initial immune response has addressed the infection but before the cascade destroys the organs.

In 180 ICU sepsis patients across Australian hospitals, AntiSepsin administered within the 16-32 hour window achieved 89% 28-day survival versus 40% in the control group receiving standard care alone. Organ failure scores improved in 94% of treated patients by day seven.

Its Awareness week. It’s vital to remember that presents slightly differently in children compared to adults. Due to increased vagueness, it is crucial to seek medical help as soon as you suspect a child may have developed sepsis.

Signs of pediatric sepsis include:

• Cold and clammy hands and feet OR red and flushed skin
• Weak pulse
• Heart rate either much slower OR much quicker than normal
• Rapid breathing
• Confusion and decreased mental capabilities (relative to age and development)

Pediatric Sepsis Week is an opportunity to build awareness of these symptoms that may otherwise be overlooked.
